贵州地区不同人群血清庚型肝炎病毒核酸检测和部分核苷酸序列分析
Analysis of nucleic acid and partial gene sequence in serum of hepatitis G virus on different population of Guizhou area
【摘要】 目的了解贵州地区庚型肝炎病毒感染状况,分析度型肝炎病毒贵州株的基因特点。方法以废型肝炎病毒(HGV)的NS3区两对寡核苷酸为引物,用逆转录套式聚合酶链反应(RT-nPCR)检测不同人群血清中庚型肝炎病毒核酸(HGV RNA,并用PCR产物直接测序分析3价阳性血清5’非编码区的部分核苷酸序列。结果60位正常人,37例血液病,86例肝病患者及107例静脉药瘾者血清HGV RNA阳性率分别为1.7%(1/60),16.22%(6/37),25.58%(22/86)和35.51%(38/107)。在肝病组中,急性非甲~戊型肝炎,慢性肝炎,肝硬化的HGV RNA阳性率分别为25.0%(3/12),23.53%(4/17)和26.32%(15/57)。测定的156个核苷酸中,3株贵州株的同源性均高于94.2%。代表株G3与国外株(U36380,U44402, U45966)同源性分别为83.9%, 90.4%和89.1%。与国内株(C964, G006)同源性分别为92.7%和96.8%。3株贵州林均存在缺失突变。结论贵州存在HGV感染,静脉药瘾者中有较高的HGV感染率。HGV可能是急性非甲~成型肝炎的病原因子。3株贵州株?
【Abstract】 Objective To investigate the prevalence of hepatitis G virus infection and analysis partial nucleoti- de sequence. Methods Two hundred and ninety sera were collected from different population in Guizhou area and HGV RNA in the sera was detected by reverse transcription nested polymerase chain reaction (RT-nPCR). The 156bp fragments from 5’NCR of HGV isolaed from the sera of three patients with non-A-E hepatitis were purified and directly sequenced. The nucleotide sequence of a representative strain (HGV-G3) was compared with those of the HGV U36380, U44402, U45966 reported in American and HGV C964, G006 reported in China. Results The positive rates of HGV RNA in 60 normal adults, 107 intravenous (IVDUS), 37 patients with homeopathy, 86 pa tients with liver diseases were 1.7% (l/60), 35.51% (38/lo7), 16.22% (6/37) and 25.58% (22/86),respec- tively. The nucleotide homology of the 5,NCR was from 94.2% to 97.4% within 3 HGV Guizhou strains (G1- G3). AS compared with published HGV sequence, the nucleotide homology between HGV-G3 and HGV U3638, U44402, U45966, HGV C964, HGV-G006 were 83.9%, 90.4%, 89. l%, 92.9% and 96.8%, respectively. The deletion of nucleotides was distributed in sequence. Conclusion There are hepatitis G virus infection in Guizhou area, the infection rate was higher in IVDUs. The nucleotide homology was higher between strains from China than that between strains from Guizhou and abroad.
