离子注入麦角甾醇酵母选育研究
A study on producing ergosterol yeast induced by ions implantation
【摘要】 用 1 0keV、剂量为 2 .6× 1 0 13 N+/cm2 ~ 8.0× 1 0 14 N+/cm2 的氮离子注入产麦角甾醇酵母 ,可产生可遗传的变异。离子注入产麦角甾醇酵母的存活率与注入剂量呈负相关 ,在存活率为 2 5 %~ 45 % ,即注入剂量为 1 .3~ 2 .3× 1 0 14 N+/cm2 时菌种有较高的正变率。最终筛选到的高产菌株YA1和YA2 ,麦角甾醇得率较出发菌株分别提高了 6 0 %和 5 5 %。经复筛及传代实验表明高产菌株遗传性能稳定。
【Abstract】 Some mutants of producing ergosterol yeast were obtained after implanted by nitrogen ions with energy 10 keV and dose from 2.6×10 13 ions/cm 2 to 8.0×10 14 ions/cm 2.The survival rate of mutants had negative relation with implantation dose.From the survival rate of 25% to 45%,ie.with the implantation dose 1.3 to 2.3×10 14 ions/cm 2,mutants had high positive mutation rates.The pure ergosterol content of mutant YA1 and YA2 was increased by 60% and 55% respectively,compared with the original strain.
