一种新的黄芩类黄酮3’,5,6’,7-四羟基-2’,8-二甲氧基黄酮的体外免疫药理作用

【摘要】 目的 :对新发现的一种黄芩类黄酮 ,3’,5 ,6 ’,7-四羟基 - 2 ’,8-二甲氧基黄酮 ( TDF)的免疫药理作用进行研究。方法 :检测 TDF对人多形核细胞 ( PMN)、单个核细胞 ( MNC)化学发光反应、化学趋向性和 PHA诱导淋巴细胞增殖的影响。结果 :TDF本身不能激活 PMN和 MNC的氧呼吸爆发 ,TDF对 PMN和 MNC无毒性作用 ;当 TDF与 PMN或 MNC预先孵育后 ,明显的抑制 f ML P或 OZ激活 PMN以及 f ML P激活 MNC的氧呼吸爆发 ,抑制的强度与孵育的剂量成正比关系 ;对PMN的抑制能力大于 MNC。 TDF本身对 PMN和 MNC没有明显的化学趋化作用 ,但经与 TDF预先与细胞孵育后的 PMN或 MNC对 ZAS的化学趋向性明显增强 ,对 f ML P的化学趋向性作用不明显 ;TDF浓度在 ( 1.2 5× 10 - 4~ 5× 10 - 4 ) mol/ L 时 ,显著地抑制 PHA诱导的淋巴细胞增殖 ,抑制率达 31%~ 10 0 %。结论 :TDF对人的免疫细胞有重要的作用

【Abstract】 Objective:To investigate immunopharmacological profile of a new plant flavonoid,3’,5,6’,7tetrahydroxy2’,8dimethoxyflavone from scutellaria baicalemsis georgi.Methods:Test the effect of new flavonoid,3’,5,6’,7tetrahydroxy2’,8dimethoxyflavone isolated from Scutellaria baicalensis georgi on chemiluminescence (CL) and chemotactic response of human ploymorphonuclear cells (PMN) or mononuclear cells (MNC) and PHAinduced proliferation of human Lymphocytes.Resuits:3’,5,6’,7tetrahydroxy2’,8dimethoxyflavone had no toxin to the PMN or MNC and did not induce CL of the PMN or MNC.When proincubated with PMN or MNC,3’,5,6’,7tetrahydroxy2’,8dimethoxyflavone strongly inhibited fMLP and opsonized zymosan (OZ)induced CL of PMN and fMLP induced CL of MNC.The inhibition was dosedependent and the inhibitory effect on PMNs was stronger than that on MNCs.3’,5,6’,7tetrahydroxy2’,8dimethoxyflavone itself had no chemotactic activity that was observed in PMNs and MNCs.However,PMN or MNC preincubated with 3’,5,6’,7tetrahydroxy2’,8dimethoxyflavone exhibited a higher chemotactic activity towards ZAS and fMLP than that towards medium at high concentration and when incubated with 3’,5,6’,7tetrahydroxy2’,8dimethoxyflavone,PMNs did not show enhanced chemotactic activity to fMLP.3’,5,6’,7tetrahydroxy2’,8dimethoxyflavone from (1.25×10 4 to 5×10 4 )mol/L concentration significantly inhibited PHAinduced lymphocyte proliferation.Inhibiting rate was 31% to 100%.Conclusion:3’,5,6’,7tetrahydroxy2’,8dimethoxyflavone has a important role in the human cells.
