三株肝癌单克隆抗体相应抗原在肝癌细胞中的表达与分布
THE EXPRESSION AND DISTRIBUTION OF THREE HUMAN HEPATOMA CARCINOMA mAbs’ANTIGENSIN HEPATOMA CELL LINES
【摘要】 应用免疫细胞化学方法、图像分析技术和免疫电镜技术 ,对 3株肝癌 m Abs A10 ,E5和 F11在肝癌细胞株中的表达情况及分布特点进行了研究。结果显示 :3株单抗抗原在肝癌细胞株 HHCC中均有较高表达 ;单抗 A10 ,F11相应抗原主要分布在肝癌细胞的胞膜和胞浆中 ,而在胞核上无明显分布。单抗 E5相应抗原除分布在胞膜和胞浆外 ,在细胞核上也有明显浓聚。本实验明确了 3株肝癌单抗相应抗原在肝癌细胞中的分布特点 ,对于深入分析抗原的本质有重要参考价值。
【Abstract】 In order to investigate the expression and distribution characteristics of three human hepatoma m Abs’ ( A1 0 ,E5,F1 1 ) antigen in hepatoma cell lines,the immunocytochemical method,image analysis and immunoelectron microscopic technique were used.The results were as follows:( 1 ) we found a hepatoma cell line HHCC which had a relatively high expression of three m Abs’antigens. ( 2 ) m Abs A1 0 and F1 1 ’s antigen was localized in cytoplasm and or cell membrane,whereas m Ab E5’santigen had a higher concentration in nucleus than in other partof hepatoma cell.These results help us to choose a suitable cell line that we will use to constructa c DNA expression library screened by the m Abs,and also give us some suggestions of the nature of the m Abs’antigens.
