变形链球菌Ⅰ型和Ⅲ型合成葡聚糖的研究

【摘要】 目的 通过对Ⅰ型JBP(c 型) 和Ⅲ型6715(g 型) 变链菌在不同状态葡糖基转移酶(GTF) 所合成的水不溶性葡聚糖(IG) 和水溶性葡聚糖(SG) 的测定来研究两种变链菌的GTF-I和GTF- S是否存在差异。方法 制备JBP和6715 的含酶上清液(cell-free enzyme) 及含酶提取液(cell- bound enzyme),以蔗糖为底物,恒温培养后分别收集IG 和SG。采用红外光谱、薄层层析及Scott 蒽酮定糖法对葡聚糖样品作定性、定量分析。结果 Ⅰ型JBP(c)含酶上清液主要合成SG,含酶提取液主要合成IG。Ⅲ型6715(g)含酶上清液主要合成IG,含酶提取液合成IG稍多于SG,但无统计学显著性差异。对于含酶上清液IG 的合成,6715 明显多于JBP(P< 0 .05) ,而SG 的合成,JBP明显多于6715( P< 0.05) 。含酶提取液IG的合成,JBP明显多于6715( P< 0.05),而SG 的合成,6715明显多于JBP(P< 0.05)。结论 变链菌Ⅰ型JBP的IG主要由细胞结合型GTF 所合成,而变链菌Ⅲ型6715 的IG主要由细胞游离型GTF所合成。这种来自不同型GTF 的IG 是否对变?

【Abstract】 Objective To study the differences of various types of GTF in Streptococcus mutans(serotype c) and Streptococcus sobrinus (serotype g) by determining water-insoluble glucans(IG) and water-soluble glucans(SG) synthesized from various glucosyltransferase. Methods The supernatant containing GTF and extraction containing GTF in these two streptococci have been prepared and glucans were synthesized. The thin layer chromatography and infrared spectra analysis were adopted to study the chemical nature and the chemical structure of glucans. Results IG presented predominantly in extraction of S.mutans and supernatant of S.sobrinus, while SG presented predominantly in supernatant of S.mutans. IG synthesized by the supernatant of S.sobrinus was more than that did by the supernatant of S.mutans, (P<0.05), whereas SG synthesized by the supernatant of S.mutans was more than that did by the supernatant of S.sobrinus (P<0.05), IG synthesized by the extraction of S.mutans was more than that did by the extraction of S.sobrinus(P<0.05). Conclusion IG was mainly synthesized by cell-bound GTF of S. mutans and SG was mainly synthesized by cell-free GTF of S.sobrinus. Further studies are required to prove the various types of GTF and whether their synthesizing IG affect streptococcus adhesion or not.
