【摘要】 目的　探讨 P16基因的突变在人胰腺癌发生及侵袭转移中的作用 ,为临床提供有益的帮助。方法　应用 PCR、PCR-SSCP、DNA测序技术 ,研究 P16基因的突变情况。结果　在 35患者胰腺癌标本中 ,发现有 12例在 P16基因第二外显子部分存在至少 5 2 2 bp的纯合缺失、另有 7例存在 2个点突变 ,总突变率为 5 4.3% ,而癌旁组织无 P16基因的突变 ,统计学分析表明 P16基因的突变与人胰腺癌多种生物学行为及临床分期 期有明确的关系。结论　研究结果提示 P16基因的突变人胰腺癌的发生、组织分化、浸润程度、淋巴结转移及临床分期有明显的相关性 ,有助于判断胰腺癌的恶性程度和患者的预后。更多还原

【Abstract】 Objictive To investigate the Relationship between genetic alteration(homozygous deletion and point mutation)of P16 Gene and Genesis,Invasion,Metastasis of Pancreatic Carcinoma.Methods Cell-cycle inhibitors were analyzed for genetic alteration and expression by Polymerase chain reaction(PCR),single strande conformation polymorhism analysis(SSCP),DNA sequencing.Result The analysis of pancreatic carcinoma for P16 gene reaveled alteration in 19 of 35 case,54.3%.12/35 pancreatic carcinoma had 522bp homozygous deletion at least and 7/35 case had two point mutation which site is same.Conclusion These result support that loss of P16 function could be involved genesis,Invasion and metastasis of panreeatic carcinoma.更多还原
