电离辐射对小鼠骨髓造血细胞周期进程的影响

【摘要】 采用流式细胞术(FCM)观察了X射线全身照射后小鼠骨髓造血细胞周期进程的时程变化和剂量效应关系,为探讨电离辐射对小鼠骨髓细胞周期进程的影响,。结果显示,2GyX射线全身照射后12h骨髓细胞出现G1期细胞百分数升高(p<0.05),4hS期细胞百分数下降(p<0.05),G2+M期细胞百分数于照后4h和12h升高(p<0.05)。量效研究显示,0.5~6Gy照射后12hG1期细胞百分数高于对照组,其中1、2、4、6Gy照射组有统计学差异(p<0.05~p<0.01),S期细胞百分数呈剂量依赖性下降(p<0.01~p<0.001),G2+M细胞百分数呈剂量依赖性增加(p<0.05~p<0.001)。提示2GyX射线全身照射后可诱导造血细胞出现G1、G2期阻滞,抑制DNA合成,并且这些变化在0.5~6Gy范围内呈剂量依赖性关系。

【Abstract】 The changes of cell cycle progression in drice bone marrow hemopoietic cell and the dose-effect celationship after X-ray whole body irradiation (WBI) were studed by flow cytometry (FCM) The data showed the elevation of the percentage of cells in G1 phase 12h after irradiation (p <0.05), the decrease of cells in S phase 4h (p <0.05) and the increase of cells in G2+M phase 4h and 12h after 2.0Gy WBI. 12h after 0.5~6.0Gy irradiation the percentage of cells in G1 phase became higher than that of sham-irradiation, especialy in 1.0, 2.0, 4.0 and 6.0Gy groups (p <0.05 ~ p <0.01), both the decrease of S phase percentage (p < 0.05 ~ p <0.01)and the increase of G2+M phase (p <0.05 ~ p<0.001) were in a dose-dependent manner. Ourresults suggested that 2.0Cy X-ray WBI to mice might lnduce the arrest in G1 and G2 phase and the suppression of DNA synthesis, these changes were all in dose- dependent manner within 0.5~6.0Gy.
