新型荧光光纤免疫磁珠流动分析系统研究(Ⅱ)
Study on a Novel Flow Fluorescence Optic Fiber Immunoassay System with Immunomagnetic Beads (Ⅱ)
【摘要】 采用荧光素异硫氰酸酯 ( FITC)对新建立的荧光光纤免疫磁珠流动分析系统参数及性能进行了考察。不同泵速下测定流量的恒定性 ,其 RSD为 0 .2 3%~0 .76 % ( n=2 4 ) ;对 1× 1 0 -5~ 1× 1 0 -2 mol·L-1FITC溶液 ,线性相关系数 0 .986~ 0 .999;连续测定 1 2次 ,RSD为 0 .73%~ 0 .95% ;2 h内基线不产生飘移。
【Abstract】 The combination of four advanced techniques,fluorescence immunoassay, optic fiber sensor , flow analysis and immunomagnetic beads separation,was developed into an analytical system. The FIA parameters were investigated in detail, and the optimum condition for the flow injection analytical system was established. The RSD is 0.23%~0.76%( n =24) for the steadiness of flow measured at different pump rates.The correlation coefficients are 0.986~0.999 for the FITC solution over 1×10 -5 ~1×10 -2 mol·L -1 .No drift occurs for at least two hours.The results show that the design of the flow injection analytical system is reasonable, and the performance reliable.
