免疫调节分子在卵巢上皮癌中的表达
Expression of immunoregulatory molecules in epithelial ovarrian carcinoma cell fines
【摘要】 目的 了解5种卵巢上皮癌(EOC)细胞系中免疫调节分子HLAⅠ、HLAⅡ、CD54、CD80)的表达,探讨EOC逃避机体免疫监视的机制。方法 采用流式细胞仪检测5种人EOC细胞系的HLAⅠ、HLAⅡ、CD54、CD80)表达;应用RT-PCR检测CD80mRNA的表达。结果 5种细胞系中仅COC1和COC2高表达HLAⅠ类分子(阳性细胞率在99%以上)。5种细胞系HLAⅡ类分子的表达均很低(1.40%~12.94%),SKOV3细胞CD54的表达较高(78.00%),COC1次之(33.25%),余者均较低。除COC2低表达CD80(6.63%)外,其余4种细胞系均不表达CD80。结论 5种EOC细胞系存在免疫调节分子表达的异常(如HLAⅠ下调、CD80的缺乏等),EOC细胞可能通过多个方面逃避机体的免疫监视。
【Abstract】 Objective To investigate the expressions of human leukocyte antigen (HLA) class Ⅰ, class Ⅱ , CD54 and CD80 in 5 epithelial ovarian carcinoma (EOC) cell lines. Methods Expressions of HLA class Ⅰ, HLA class Ⅱ, CD54 and CD80 in 5 EOC cell lines were detected by flowcytometric analysis. The expressions of CD80 mRNA were determined by reverse transcription-porymerasee chain reaction (RT-PCR). Results Flow cytometric analysis showed that only COC1 cells and COC2 cells expressed HLA class Ⅰ with a high positive rate (more than 99%). There was mild degree of down regulation of HLA class Ⅰ on TYK cells. The expressions of HLA Ⅱ of all 5 cell lines were very low (from 1.40% to 12.94%). 78.00% of SKOV3 cells and 33.25% COC1 cells expressed CD54. Expressions of CD54 on the other 3 cell lines were very low. There were no expression of CD80 on 3AO, SKOV3, TYK and COC1 cells. COC2 cells expressed CD80 on the cell surface with a low positive rate (6.63%). RT- PCR analysis snowed only COC2 cells expressed CD80 mRNA, which was in accordance with flow cytometric results. Conclusions Abnormal expressions of immune regulatory molecules on 5 EOC cell lines such as down-regulation of HLA I , deficiency of CD80 may serve as the mechanisms that EOC cells can escape from immune survilance of the host.
