甘薯品种金山471凝集素的提取及其性质研究
Purification and characterization of a lectin from Jingshan 471 variety of sweetpotato
【摘要】 甘薯凝集素 (sweetpotato lectin,简称 SPL )由甘薯品种金山 4 71的叶片组织经 (NH4 ) 2 SO4 分级沉淀 ,甲壳素亲和柱及 Sephadex G- 75分离得到的 .经 PAGE或 SDS- PAGE均呈现一蛋白区带 ,用 SDS- PAGE测得亚基相对分子质量为 4 1× 10 3左右 .SPL能凝集人各种血型及部分动物红细胞 ,其中对鸽红细胞凝集活性最高 ,能凝集小鼠 S180 肉瘤细胞 .其凝集活性可被阿拉伯糖或 N-乙酰葡萄糖胺等抑制 ,在 90℃加热 2 0 min才完全失活 .SPL中性糖含量为 8.4 2 % .氨基酸组成分析表明分子中富含酸性氨基酸 .
【Abstract】 Sweetpotato Lectin(SPL) was isolated from lamima textus of Jinshan 471, a variety of Ipomoea batatas Poir, by ammonium sulfate fractional precipitation, chitin affinity chromatography column and sephadex G 75. By PAGE or SDS PAGE, it presented to be a protein belt. Subunit relative molecular mass measured by SDS PAGE is about 41×10 3. SPL can agglutinate various human blood types and some zoic akaryovyte, among these, the agglutination activity of SPL in pigeon akaryocyte is the highest. SPL can agglutinate S 180 sarcoma cell of mice. The agglutinative activity of SPL can be suppressed by arabinose, GluNAe, etc. After heating for 20 minutes at 90 ℃, SPL can be entirely devitalized. The neutro saccharide content of SPL is 8.42%. The amido acid composition analysis suggested that SPL molecules abundantly contained acid amido acid.
