运用RAPD法识别杨树品种技术的研究

【摘要】在正确提取ＤＮＡ后，探讨了在成本最低、效果最好的基础上，得出了ＤＮＡ最佳增殖反应液的组成（１０Ｘｂｕｆｆｅｒ１．５０、ｄｎｔｐｓ０．３０、ＭｇＣｌ２０．６０、灭菌水１０．５７、Ｐｒｉｍｅｒ１．００、Ｔａｇ０．０３、ＤＮＡ１．００），其次用６０个引物对杨柳科属和杨属的１３个品种进行了人工分析，发现多态标记３２个，用其中的任意１３个标记就能区别１３品种。更多还原

【Abstract】 The best reaction composition have been studied in this paper. It is that 10xbuffer 1. 50μl,dNTP 0. 30μl,MgCl2 1. 60μl,h20 10. 57μl,primer 1. 00 μl,TaqDNA 0. 03μl.Template DNA 1. 00μl. The results show htat: 32 polymorphic markers have been found when 60 primers were used to identify the 13 varieties of Populus,and 13 arbitrary markers can identify them completely.更多还原
