C6脑胶质瘤动物模型肿瘤细胞的超微结构观察
Observation of ultrastructure for specimens of C6 glioma animal model and juddgement of malignant degrees
【摘要】 本文对C6脑胶质瘤动物模型肿瘤标本进行了超微结构观察,以判定肿瘤生长方式及恶性程度。取成年雄性Wistar大鼠作为实验对象,采用立体定向方法将对数生长期的C6胶质瘤细胞悬液(1×106ml)注入靶点。2~3周后手术切除肿瘤并进行电镜标本制备,H600A型透射电镜观察。观察结果显示:肿瘤中心区有两种肿瘤细胞核型;不规则核型大于55%,细胞浆结构发育较差,核浆比例明显大于1。肿瘤与正常脑组织无明显边界,亦无包膜,呈浸润型生长,病理分级约有Grade~(WHO1993)。我们认为:C6细胞脑胶质瘤动物模型超微结构的观察,对进一步研究人类脑胶质瘤的恶性程度,预后判定及综合治疗具有指导意义
【Abstract】 Objective: To made the observation of ultrastructure for specimens of C6 glioma animal model in order to judge the way of growing of tumor and the malignancy of degree. Methods:Experimental animals were grown\|up pure male Wistar rats. The suspension of C6 tumor cells (1×10\+6/ml exponential growth) were injected into the targeting point by means of stereotaxic instrument. The tumor specimens were resected and were ready to use for H\|600A electron microscope samples after 2\|3 weeks. Results:Through the observation of the central and marginal area of the tumor under the electron microscope, two kinds of nuclei were found in the C6 cells of tumor model. One of them was extremely irregular, the irregular nucleus took about more then 55%. The structure of cyotplasm of C6 cells were poorly developed with the proportion of nucleus\|cytoplasm maginficantly bigger than 1. No clear borderline of existence of tumor limiting membrance were found between normal brain tissee uner electronic microscope. The pathological diagnosis was astrocytoma grade Ⅱ\|Ⅲ (WHO 1993).Conclusion:The observation of ultrastructure for specimens of C6 glioma animal model is significant for the studies of human being glioma, especially to research the malignancy of degree, the judgement of prognosis and mutitherapy of glioma.
