氟对大鼠胚胎卵黄囊损害作用的时间──效应关系

【摘要】 应用大鼠植入后全胚胎培养,在氟离子浓度为30μg/ml于胚胎培养的不同时间段染毒,观察卵黄囊直径、血管分化形成及超微结构的改变。结果表明,氟作用16h,开始对卵黄囊血管分比和血管网形成有明显抑制作用;作用24h以上,卵黄囊直径明显减小,同时卵黄囊超微结构如细胞表面微绒毛,以及胞内线粒体等细胞器发生了明显的病理改变,对营养的摄取功能降低。染毒时间越长上述变化越严重。提示氟对卵黄囊结构和功能的损害可能是其对啮齿类动物胚胎毒性作用的重要环节之

【Abstract】 Embryoes of rat on day 9.5 of gestation were explanted and cultured with exposure to fluoride 30ug/ml rat serum for 8,16,32 and 48 hrs respectively. After 48 hrs in culture, development of yolk sac its ultra- structures of each viable embryo were observed. The results showed that the differentiation of blood vessels in yolk sac of 16 hrs exposed to fluoride was inhibited obviously. After 24,32 and 48 hrs exposure, resulted in relevant decrease in yolk sac diameter, the formation of blood vessel network, as well as changes of the ultrastructures(decrease in the number of microvilli structure, arragement in disorder or falling off; a portion of mitochondria swelling and reduction in the number of critae,etc)and reduced phagocytosis of yolk sac cell.
