pSVPoM cat微基因修饰雪旺细胞在鼠脊髓内存活及基因表达
Long-term survival and gene expression of intraspinal implantation of pSVPoMcat genetically modified Schwann cells in rats
【摘要】 目的 观察 p SVPo Mcat微基因修饰雪旺细胞 (SC)在鼠脊髓内存活及基因表达 .方法 实验动物分为 3组 ,A组 p SVPo Mcat微基因修饰 SC移植组 ;B组高纯化 SC移植组 ;C组为脊髓损伤 (SCI)对照组 .术后动物分组存活 2 ,4,8及 12 wk(每组每次 2 0只 ) ,取移植区脊髓切片 ,进行 S- 10 0蛋白 ,髓鞘碱性蛋白 (MBP)免疫细胞化学染色及地高辛 (DIG)标记的 h MBE2 c DNA探针的原位杂交 (ISH)测定 .结果 术后 2 ,4,8及 12 wk,A组的 S- 10 0和 MBP染色细胞以及ISH细胞分别为 ,(82 .3± 6 .0 7) % / (77.8± 12 .3) % / (6 7.8±8.6 ) % ;(81.2± 5 .2 ) % / (76 .3± 11.8) % / (6 8.4± 8.3) % ;(80 .7± 5 .6 ) % / (75 .4± 11.6 ) % / (6 6 .4± 8.2 ) % ;(81.6±6 .2 ) % / (75 .8± 9.5 ) % / (6 3.3± 8.0 2 ) % (P>0 .0 5 ) .其中ISH阳性细胞可见髓鞘形成 ,B组术后 2 ,4,8及 12 wk的 S-10 0染色的细胞为 ,(6 6 .3± 6 .8) % ,(4 0 .2± 11.2 ) % ,(10 .8± 2 .6 ) % ,(3.2± 0 .6 ) % (P<0 .0 0 1) .未发现 MBP染色细胞以及 ISH细胞、C组未发现任何阳性细胞 .结论 p SVPo Mcat微基因修饰 SC在鼠损伤脊髓内能长期存活并表达外源基因 ,且有助于受伤脊髓的髓鞘形成 .它们为脊髓损伤的基因治疗提供了可行途径
【Abstract】 AIM To observe the long term survival and gene expression of genetically modified Schwann cells (SC) with pSVPoMcat in the spinal cord injured, the cells were implanted into the spinal cord. METHODS The experimental animals were divided into three groups, randomly: pSVPoMcat modified SC implantation group (group A), SC implantation group (group B), control group (group C). After three months, the sections of spinal cord grafted were checked by immunocytochemistry and in situ hybridization in three groups. RESULTS There were a large staining positive cells of S 100, MBP and positive ISH cells in group A. The myelin of positive ISH cells rounded new regeneration axons. There were a few positive cells of MBP in group B, but no positive cells in group C. CONCLUSION pSVPoMcat genetically modified SC has a long term survival in intraspinal injury and helps to form myelin of the spinal cord injured.
