大鼠头颅瞬间侧向旋转致伤后脑微血管的超微结构特征

【摘要】 目的 研究大鼠头颅瞬间侧向旋转后脑微血管超微结构特征 .方法 大鼠 12只等分为对照、损伤 (伤后 2 h观察 )及 (伤后 2 4h观察 )组 .制头颅瞬间侧向旋转脑损伤模型 .每组 2只常规灌注固定后行延髓组织透射电镜观察 ,另 2只灌注多聚树脂预聚体 ,取延髓组织盐酸消化 ,选血管铸型团行扫描电镜观察 .结果 大鼠头颅旋转后 2~ 2 4h,延髓切片透射及血管铸型扫描电镜下均见毛细血管痉挛、狭窄 ,形成微血栓及微绒毛、内皮细胞变性 ,血管周围胶质足突肿胀 ,管外有水肿间隙 ,管壁不全或完全断裂 ,出现无血管区 .结论 头颅瞬间侧向旋转可致脑微血管结构损害 .脑微循环异常是DAI重要病理生理变化 .改善脑微循环有利于 DAI临床治疗

【Abstract】 AIM To study the ultrastructural characteristics of cerebral microvessels of rats due to instant lateral head rotation. METHODS Twelve rats were equally divided into sham group, injury group (observed at 2 h after injury) and group Ⅱ (observed at 24 h after injury). The rat models of brain injury were produced by instant lateral head rotation. Two rats in each group were fixed by perfusion and the tissues cut from the medulla oblongata were prepared for transmission electron microscopic observation. The other two rats were perfused by performed polymer of polyresin and the tissues of medulla oblongata were digested in hydrochloric acid and the vascular casts were selected and prepared for scanning electron microscopic observation. RESULTS At 2~24 h after lateral rotation of rat head, it was found, under transmission and scanning electron microscope, that the tissue sections and vascular casts from medulla oblongata showed evidence such as capillary spasm and narrowing, formation of microthrombi and microvilli, endothelial degeneration, perivascular glial process swelling, occurrence of extravascular edema, partial or total disconnection of microvessels, and presence of vessel disappeared areas. CONCLUSION Instant lateral head rotation is capable of leading to the damage of cerebral microvessels in structure. The abnormality of brain microcirculation is an important pathophysiological change in DAI. It is beneficial to improve brain microcirculation for clinical therapy of DAI.
