入肝血流阻断对正常大鼠部分肝切除术后肝再生的影响
Effects of occlusion of blood flow into liver on liver regeneration after partial hepatectomy in rats
【摘要】 目的 探讨入肝血流阻断对部分肝切除术后正常大鼠肝再生的影响。方法 健康雄性Wistar大鼠随机分为 3组 :PTC组 ,肝门阻断 15min切肝组 ;PH组 ,部分肝切除组 ;SO组 ,假手术组。测定术后肝脏重量的变化 ;用免疫组化法检测术后 2 4、4 8、72h和 7d各时相点PCNA标记指数 ;观察各组术后血清前白蛋白 (PA)的变化。结果 PTC组较PH组术后肝再生率下降 ;PCNA标记指数降低 ,高峰延迟 ;PA水平恢复缓慢 ;术后动物死亡率增加。结论 阻断大鼠第一肝门 15min可致肝细胞功能受损 ,肝再生延迟。
【Abstract】 Objective To study the effects of occlusion of the portal vein and hepatic artery on the liver regeneration after partial hepatectomy in rats. Methods Male Wistar rats were randomized into 3 groups; group PTC, in which the rats received occlusion of the portal vein and hepatic artery at hepatic hilum for 15 minutes and partial hepatectomy was performed, group PH, in which the rats were operated on for partial hepatectomy and group SO in which the rats received a pseudo operation.The labeling index of proliferating cell nuclear antigen(PCNA)was measured on the 1st, 2nd, 3rd and 7th day after operation. The liver weight was recorded at different time points postopertively to assess the rate of liver regeneration. The serum level of prealbumin was determined.Results In the group PTC,the rate of liver regeneration was slower than that of group PH, the index of PCNA was decreased with a delayed peak and the serum level of prealbumin was slowly recovered. In addition, the highest mortality rate occurred in group PTC.Conclusion 15 minutes of the occlusion of the portal vein and hepatic artery before partial hepatectomy damages liver functions and delays liver regeneration in rats.
