硒对培养状态下大鼠大脑皮层神经元存活及早期突起生长的影响
The Influence of Selenium on Survival and Outgrowth of Processes of Newborn Rats’ Cerebral Cortex Neurons
【摘要】 探讨硒对大鼠大脑皮层神经元存活及突起生长的影响 ,利用新生 SD大鼠大脑皮层神经元体外分散培养技术 ,培养液中加入不同浓度的硒 ( 6 2 .5 μg/ L,12 5 .0 μg/ L,182 .5 μg/ L)并以神经生长因子 ( NGF)作阳性对照 ,测量了 16 h、2 4h、36 h、48h小时后神经元最长突起长度及平均每视野神经元存活数 ( 3d,5 d,7d)。结果显示 ,低浓度的硒与空白对照组相比可见加硒组存活数增加 ,突起生长早且长。低浓度硒对大脑皮层神经元存活及突起生长有促进作用
【Abstract】 In order to investigate the effect of selenium (Se) on survival and outgrowth of processes of neurons from cerebral cortex in vitro, we also observed control group, the nerve growth factor (NGF) as the positive group by using the technique of neuron primary culture with addition of three different dose and low concentration selenium, at the same time. We investigated the number of survival neurons on the 3d、 5d、 7d and the outgrowth processes of neurons in 16~24h, 36~48h. Results showed that selenium could increase both in the number of survival neuron and in the length outgrowth of neurite. The development of neurite is more early than those of the control group.
