发根农杆菌介导的抗病毒基因导入番茄转化系统的建立
Establishment of agrobacterium rhizogenesis mediated transformed system on transferring virus resistant genes into tomato
【摘要】 试验设置基因型、菌液浓度、外植体和方法 4个因素 ,利用发根农杆菌将抗 TMV (烟草花叶病毒 )的 TMV- CP基因和抗 CMV (黄瓜花叶病毒 )的 CMV- CP基因按 L16( 4 4× 2 3)正交表转化番茄。 Km抗性愈伤组织频率的结果表明 ,不同品种、不同菌液浓度转化频率存在着显著性差异 ,而外值体部位、方法之间差异不显著。并已得到 9株转化再生的番茄植株 ,经 PCR检测和EL ISA检测证明 TMV- CP和 CMV- CP基因已导入番茄并表达
【Abstract】 Four factors,genotype,the densities of bacterial suspension,explants and transformed methods,were set up in this experiment.Target genes,TMV CP gene and CMV CP gene,were transferred into tomatoes according to orthogonal table L 16 (4 4×2 3)by agrobacterium rhizogenesis 。The results of Km resistant calli’s frequency demonstrate:Significant differences exist between genotypes and between different densities of bacterial suspension,and Significant differences don’t exist between explants and between different methods.9 transgenic tomatoes were gained.Through PCR and ELISA detection,TMV CP gene and CMV CP gene were proved to be transferred into tomatoes and expressed in the part of transgenic tomatoes.$$$$
