转促凋亡基因胃癌细胞的光动力学治疗实验研究
A study of photodynamic therapy for the gastric adenocarcinoma cell line transduced by the genes relating to apoptosis
【摘要】 评价转促凋亡基因胃癌细胞的光动力学治疗效果。方法 :分别用携带野生型 p5 3cDNA和反义bcl 2序列的逆转录病毒转导胃腺癌细胞系MGC80 3 ,稳定表达后加光敏剂并红光照射 ,检测细胞存活及凋亡。结果 :在光敏剂及红光作用下野生型 p5 3及反义bcl 2都能使转导细胞的存活率降低 ,凋亡增加。 结论 :野生型 p5 3及反义bcl 2可增加光敏剂 2 丁胺 2 去甲氧基竹红菌甲素对细胞的光敏毒性。细胞对光敏剂诱导凋亡的敏感性明显增高。推测该光敏剂的光动力治疗途径可能以诱导细胞凋亡为主。
【Abstract】 Objective: To estimate the efficiency of transferring genes relating to apoptosis combined with photodynamic therapy for the gastric adenocarcinoma. Methods: Transduction of the retrovirus vectors carrying wild type p53cDNA and antibcl 2 sequence to a gastric adenocarcinoma cell line MGC803 was carried out respectively. After screening by G418, the photosensitizer 2 butylamino 2 demethoxy hypocrellin A (2 BA 2 DMHA) and the red light were administered. Then the survival rate of the cells and their apoptosis were detected. Results: Under the condition of 2 BA 2 DMHA and red light, wild type p53 and antibcl 2 could reduce survival rate and increase apoptosis. Conclusion: Both wild type p53 and antibcl 2 may enhance the phototoxicity of 2 BA 2 DMHA and promote the apoptosis in MGC803 cells. The main photodynamic role of 2 BA 2 DMHA in the tumor cells may be to induce apoptosis.
