污染稻麦籽实中镉和铅的分布及其存在形态
DISTRIBUTION AND EXISTING FORMS OF CADMIUM AND LEAD IN POLLUTED SEEDS OF RICE AND WHEAT
【摘要】 组织化学及生物化学分析表明 :污染稻麦籽实中Cd和Pb呈不均一分布 ,胚中质量含量显著高于胚乳 ,且皮层和颖壳中含量也较高 ,但胚乳中的蓄积总量最多 ;在籽实主要营养成分中Cd和Pb以与蛋白质相结合的形态为主 ,其中与谷蛋白、球蛋白的结合量最多 .葡聚糖凝胶层析分析表明 ,稻米与麦粒中的Cd和Pb主要与蛋白质结合 ,其结合的表观相对分子质量分别约为54.50× 10 3和 5.50× 10 3.随着样品加工深度的升级 ,稻和麦籽实中这 2种元素的去除率增高 ,水稻中Cd和Pb去除率分别为 2 4 .1%与 56 .9% ,小麦分别为 6 1.6 6 %与 81.2 7% .
【Abstract】 The distribution of cadmium and lead in polluted seeds of rice and wheat and the binding forms of heavy metals are studied through histochemistry and biochemistry methods. The results show that the distribution of Cd and Pb in the seeds is uneven. The concentration of Cd and Pb in embryo is the highest among the various components of polluted seed. The concentration of Cd and Pb in the chaff and seed capsule is the also relatively high. The accumulating contents of Cd and Pb in endosperm are predominant. Cd and Pb in various nourish components of crop seeds are mainly existing in the protein binding forms, which are mainly bound with globular and glutelin. The results of gel chromatograph (Sephadex G75) show that Cd and Pb are mainly bound with protein in rice and wheat. The relative molecular mass of Cd, Pb protein is about 54.50×10 3 and 5.50×10 3 respectively. The removal ratio of Cd and Pb in rice is 24.10 %, 56.93% respectively and that in wheat is 61.66 %, 81.27% respectively.
