牦牛血超氧化物歧化酶电泳多谱带现象研究

【摘要】 采用聚丙烯酰胺凝胶电泳、SDS 聚丙烯酰胺凝胶电泳、线性梯度聚丙烯酰胺凝胶电泳、制备电泳、等电聚焦电泳以及不同试剂处理等多种方法对牦牛血铜锌超氧化物歧化酶电泳多谱带现象进行了研究。结果表明在不同的实验条件下 ,各带会发生相互转化。该多谱带现象是由聚合和构象异构体两种原因造成

【Abstract】 In the research of SOD, we always found that the SOD which was purified to homogeneity gave not only one band in polyacrylamide gel electrophoresis and all bands showed SOD activity. It has been found that this polymorphism of SOD was changed under different experimental conditions, which showed that the polymorphism was not caused by the difference of genes or the difference of length or side groups of the peptide chains which was caused during the processing after transcript or the purification. According to the references about the protein research, we estimated that the possible reasons of the polymorphism were polymerization, conformational isomers and charge isomers. There was some research on the SOD charge isomers, but not on this polymorphism. In this thesis, we have done some research on the electrophoresis polymorphism of Cu·ZnSOD from yak erythrocyte by continuous gradient polyacrylamide gel electrophoresis, preparative electrophoresis, electric focusing electrophoresis and dealing with some different reagents. Our conclusion was that the polymorphism of the Cu·Zn SOD from yak erythrocyte was caused by polymerization and conformational isomers. We think this research can give some references on the researches of physiology, genetics, developmental biology, evolution and taxonomy in which the isoenzyme electrophoresis is used.
