子宫颈上皮内瘤样病变和子宫颈鳞癌组织中p53与MDM2的表达及其与PCNA的关系
The expression of p53, MDM2 and their associations with proliferating cell nuclear antigen in cervical intra-epithelial neoplasm and cervical carcinoma
【摘要】 目的:探讨MDM2、p53蛋白、增殖细胞核抗原(proliferaingcellnuclearantigen,PCNA)在宫颈癌发生、发展中的作用。方法:用免疫组化SP法检测MDM2、p53蛋白及PCNA在76例正常宫颈上皮、宫颈上皮内瘤样病变(cervicalintra-epithelialneoplasm,CIN)、宫颈鳞状细胞癌(squamous-cellcarcinoma,SCC)组织中的表达。结果:1.随着宫颈病变加重,MDM2、p53、PCNA表达率均逐渐增高,三者在正常宫颈、CIN以及SCC组织中的表达率分别为:MDM2的表达为0、13.64%、44.12%;p53的表达为5.00%、18.18%、47.06%;PCNA的表达为5.00%、59.09%、85.29%。MDM2、p53在浸润癌组较CIN及正常宫颈组明显增高。2.MDM2、p53与PCNA的表达相关。3.MDM2的表达与p53的聚积在统计学上未显示出相关性。结论:1.MDM2、p53的异常表达可能与宫颈癌的发生、发展有关。2.癌前病变及癌组织中细胞增殖活性明显增高。
【Abstract】 Objective: To investigate the roles of MDM2, p53 protein and proliferating cell nuclear antigen (PCNA ) in the pathogenesis and development of cervical carcinoma. Methods: We detected the expression of MDM2, p53 protein and PCNA in normal cervical epithelium, cervical intra-epithelial neoplasm (CIN ) and invasive cervical squamous-cell carcinoma(SCC) with immunohistochemical SP method in 76 cases. Results: 1. The expression rates of MDM2, p53 ahd PCNA were gradually increased with the progress cervical lessions. The positive rates of MDM2, p53 and PCNA in normal cervical epithelium, CIN and invasive carcinoma were 0, 13. 64%, 44. 12%; 5. 00%, 18. 18%,47. 06%; and 5. 00%, 59. 09%, 85. 29%, respectively. Fhe positive rates of MDM2 and p53 in invasive cervical carcinoma group were obviously higher than those in normal cervical epithelium group and CIN group. 2. The expression of MDM2 and p53 were positively related with that of PCNA. 3. The frequence of MDM2 did not show significant correlation with accumulation of p53. Conclusions: 1. The abnormal expression of MDM2 and p53 may be involved in the pathogenesis and development of cervical careinoma. 2. The activation of cell proliferation is obviously higher in precancerous lesions and cancerous tissues.
