刺五加抗癌作用的实验研究

【摘要】 目的 :为进一步探讨刺五加对实验性肝癌发病的干预作用及其可能机理。方法 :43只雄性大鼠被随机分成 3组①正常对照组 :喂普通饲料 ② 3′-MeDAB组 :喂含 0 0 6 % 3′-MeDAB饲料 10周 ③刺五加组 :除致癌剂外另加入刺五加 (4 .5g/kg饲料 ) ,历时 2 0周。实验过程中所有动物均自由进食及饮水。 结果 :刺五加组较 3′ -MeDAB组 :①外周血液及肝组织中PAI- 1含量明显减少 (P <0 0 5 )。②肝细胞周期时相G0 /G1比率增加 (P <0 0 1) ,S期和G2 M期比率减少 (P <0 0 1) ,PI和DI减低 (均P <0 0 1) ,③肝细胞rasp2 1荧光指数减低 (P <0 0 5 ) ,④免疫组化PCNA阳性表达率降低。⑤光镜及电镜检测均显示有一定程度减轻。结论 :上述结果提示刺五加适于被开发为常规抗癌辅助药物

【Abstract】 Purpose: In order to study the role of anticancer and its mechanism of acanthopanax sentiocosus(APS) in rat liver during 3′-MeDAB hepato carcinogenesis. Methods: 43 male SD rats were randomly divided into three groups: group 1.15 rats received the basal diet as mormal control. Group 2.18 rats received 0.06% 3′ MeDAB in there diet for 10 consecutive weeks and then a basal diet. Group 3.10 rats received 0.06% 3′ MeDAB along with APS (4.5g/kg diet) daily in food for 10 weeks. And then a basal diet along with the same dose of APS till the end of experiment. All animals received the diet and water adlibitum. Results:Comparision between two groups was made and the results showed that(1) the content of plasminogen activator inhibitor l(PAI-1) of serum and liver of APS group rats was decreased significantly than 3′ MeDAB group (P<0.05),(2) The phases of the cell cycle of liver G0/G phase was increased (P<0.01), proliferation index (PI) and DNA index (DI) and the heteroploid were decreased (P<0.01), Sphase and G2M phase of APS group was decreased significantly than 3′ MeDAB group (P<0.05), (3) ras p 21 gene protein by FCM was decreased of APS group (P<0.05), (4) S P immunohistochemical localization was expression of PCNA and bcl 2. The amount of PCNA positive cell were little of APS group. There was no obvious difference of bcl 2 expression between two groups, (5) The pathological changes by light and electron microscopic observation were light. Conclusion: The present data suggest that APS exhibited a protective effects in the appearance of cancer induced by 3′ MeDAB, through inhibiting the growth of cell decreasing the expression of ras P21, decreasing the content of PAI l during hepatocarcinogenesis.
