H2O2对HL-60细胞DNA中8-羟基鸟嘌呤含量影响的研究
STUDY OF EFFECT OF H2O2 ON CONTENT OF 8-HYDROXYGUANINES IN HL-60 CELLULAR DNA
【摘要】 目的与方法 :本文采用气相色谱 /氢火焰离子检测器 (GC/FID)和气相色谱 -质谱仪选择性离子检测器 (CGC/MS -SIM )探讨H2 O2 致HL - 6 0细胞氧化损伤时DNA中氧化修饰碱基 8-羟基鸟嘌呤 (8-oh -G)含量的变化 ,观察HL - 6 0细胞脂质过氧化程度和细胞内还原型谷胱甘肽 /氧化型谷胱甘肽 (GSH/GSSG)比值的变化。结果 :结果显示H2 O2 可使HL - 6 0细胞丙二醛 (MDA)含量增高 (P <0 .0 1) ,GSH/GSSG比值显著下降 (P <0 .0 1)。细胞经H2 O2 持续作用 8h可检测到DNA中有8-羟基鸟嘌呤的存在 ,H2 O2 的浓度影响 8-羟基鸟嘌呤的生成量。结论 :结果提示 ,HL - 6 0细胞脂质化水平的升高、GSH/GSSG比值下降、细胞DNA中 8-羟基鸟嘌呤含量变化和H2 O2 浓度及作用时间有关。
【Abstract】 Purpose and Methods: The 8-hydroxyguanine is one of oxidized productions in DNA . In this report, the HL-60 cells were treated with varying concentration H\-2O\-2 and different time. Content of 8-hydroxyguanines in DNA was detected by GC/FID, CGC/MS-SIM,yields of MDA and ratio of GSH/GSSG were measured in HL-60 cells. Results: The results showed that yields of MDA was increased (P<0.01), ratio of GSH/GSSG was decreased (P<0.01) in HL-60 cells damaged by H\-2O\-2. Detection of GC/FID and GC/MS-SIM indicated the 8-hydroxyguanines be produced in HL-60 cellular DNA. Conclusion: The experiments suggested yields of MDA, ratio of GSH/GSSG and content of 8-hydroxyguanines depend on the concentration of H\-2O\-2 and treatment time.
