肝与肝癌基因表达谱差异的研究
DIFFERENCE OF GENE EXPRESSION PROFILE IN NORMAL LIVER AND HEPATOMA
【摘要】 筛选肝癌与正常肝基因表达差异克隆。方法应用Microarray膜片分别与肝癌和正常肝组织mRNA进行杂交,比较杂交信号,获得表达差异克隆。结果在正常肝中共有990个基因表达,其中255个较高表达。425个cD┐NA克隆在正常肝中的表达高于肝癌,其中33个有较大差异。190个cDNA克隆在肝癌中的表达高于正常肝,其中60个有较大差异。在这些差异克隆中,鉴定出有些是已被他人证实在其它癌中有差异的已知基因。结论应用大规模筛选表达差异的Microarray方法,获得了肝与肝癌表达差异的基因克隆。
【Abstract】 Objective:To screen for the gene clones with differential expression in normal liver and hepatoma.Methods:By means of hybridization of genes on microarray membrane with normal liver mRNA or hepatoma mRNA respectively,and by comparing hybridization signals on membrane genes,the differential expression genes were identified.Results:990 out of 18 376 cDNA clones on membrane expressed in normal liver, and 255 expressed in higher level.425 cDNA clones expressed in higher level in normal liver than in hepatoma,and 33 of them showed obvious difference.The expression of 190 cNDA clones in hepatoma was higher than in normal liver,60 of then showed marked difference. Among these differential expression genes,some of them had been identified by others as known differential genes in other cnacers.Conclusion:The differential expressed clones among normal liver and hepatoma were obtained by microarray method.
