全蝎诱导HL-60细胞程序性死亡

【摘要】 应用细胞培养和流式细胞仪分析技术分析全蝎水煎液对HL60细胞增殖、细胞形态及细胞凋亡的影响。结果全蝎水提物对HL60细胞增殖有一定的抑制作用,细胞凋亡比例由对照组的736%增加至947%(4mg/ml)、1256%(8mg/ml)和2144%(12mg/ml)。细胞形态表现为细胞增大,核膜皱缩,核固缩或核碎裂,但细胞膜完整。表明全蝎可诱导HL60细胞凋亡,并对细胞增殖有一定的抑制作用

【Abstract】 In order to study the effect scorpion on the proliferation and induction of programmed cell death(PCD) of HL 60 cells,contral and experimental groups were established.Both of them all pass the examination of FACS after 3 days′ process with 3,6,10mg/ml of scorpion.Results showed that the rate of PCD of contral was 7.36%,and the rate of experimental groups were 9.47%(3mg/ml),12.56%(6mg/ml) and 21.44%(10mg/ml).Some changes in the morphology of experimental groups included:karyopyknosis,shrinkage of the cell membrane and karyorrhexis,and contral groups had no such changes.The inhibition of scorpion on the proliferation of HL 60 cells was also proved and showed definite dose effect relationship.
