补肾醒脑解毒方对单纯疱疹病毒感染神经元的保护作用
The Protective Effects of Bushenxingnaojiedufang on Cultured Hippocampal Neurons Infected with Herpes Simplex Virus Type 1
【摘要】 目的 观察补肾醒脑解毒中药复方对 H S V- 1感染神经元的保护作用。方法 用 H S V- 1感染原代培养大鼠海马神经元造成急性感染与潜伏感染,然后检测中药煎液对神经元细胞病变及子代病毒滴度的影响。结果 培养神经元感染01 M O I H S V- 1后24 h 出现病变,6d 后70% 的细胞溶解死亡。补肾醒脑解毒方对低感染量病毒(005 M O I)所致细胞病变有较强的抑制作用,并能降低子代病毒滴度。对潜伏感染神经元能维持其正常形态10周以上。结论 补肾醒脑解毒方能维持潜伏感染神经元正常形态,并能减轻 H S V- 1所致神经元急性溶细胞病变。
【Abstract】 Objective:Observing the effects of Bushen Xingnao Jiedu Fang(BSXNJDF) on protecting cultured hippocampal neurons infected with herpes simplex virus type 1(HSV 1) in vitro.Experimental Methods:Detecting the cytopathic effect (CPE) under microscope and viral yield by means of Reed Muench’s method in cultured neurons which were treated with decoction of BSXNJDF before or after absorption of HSV 1.Observing the morphology of the neurons with HSV 1 latent infection after treatment of BSXNJDF.Results:(1)The CPE of cultured neurons was appeared at 24 hours after absorption of HSV 1 with 0.1 multiplicity of infection (MOI),and most neurons were lytic and dead at 4 days after 0.1 MOI HSV 1 infection.The CPE of cultured neurons infected with 0.05, 0.10 MOI HSV 1 was prevented in the presence of 1∶40,1∶80,and 1∶160 concentration of the decoction of BXSNJDF,when the decoction was added before 24 hours of HSV 1 absorption,and the viral yield was significantly reduced.But when the decoction was added after 0.5 hours of viral absorption,the CPE of neurons only infected with 0.01 and 0.05 MOI HSV 1 could be inhibited in the presence of 1∶40,1∶80 concentration of the decoction.(2)When the latent state of HSV 1 in cultured neurons was produced in the presence of 50uM acyclovir,the CPE of infected neurons with 0.1 MOI HSV 1 could be completely prevented for 10 weeks at the concentrations of 1∶40~1∶320 of the decoction after remove of acyclovir.Conclusion:The BSXNJDF was found to have the effects on maintaining survival and normal morphology of cultured neurons harboring HSV 1 latent infection,and reduced the CPE of neurons infected with HSV 1 and viral yield.\;
