近视与视网膜多巴胺受体放射自显影研究
Autoradiography on correlation of dopamine receptor and myopia
【摘要】 目的:通过放射自显影方法对比研究鸡视网膜多巴胺受体在近视及正常状态下的分布。方法:取正常鸡和实验性近视鸡眼球,去角膜及眼内容物后速冻,做12μp冷冻切片,分三组:[3H]SCH233切标记D1组,[3H]Spiperone标记D2受体组,非特异性竞争组,行放射自显影研究。结果:鸡视网膜D1、D2受体分布不同,D2受体分布有明显的区域性,D1不明显Z近视状态下D1、D2受体数目增多,D2增多更明显。结论:视网膜多巴胺受体两种化型均参与近视的调控,数目增多与近视视网膜多巴胶生物水平下降有关。
【Abstract】 To investigate dopaminergic mechanism on myopia by means of autoradiopphy (ARG), To studydistributions of dopamine(DA) D1 and D2 receptors in chicken retina in vitro and to compare DA receptors in myopic andnormal chicks. Methods:[3H] SCH 23390, D1 receptor ediolabeled ligand:; [ 3H] Spuperone, D2 receptor ligand, (+ ) - buta-clamol, nonspecific binding The fresh eyecups of myopic (Lid sutrued) and normal chicks were frozen on dry ice. The 12mthick tissue sechons were taken, incubaed, washed and dried. ARG expeare thek 6 ~ l2 weeks for [3HJ SCH 23390 and 1~ 6 weeks for [3H] Spiperone Results:Specilic [3H]SCH 23390 binding (D1 ) was found over all the retina, the differenceof local distribution was not obvious. SpeCilic [3H] Spiperone binding (fy ) was distributed tluDughout the retina with veryhigll density over Photoreceptor and retinal pigment epitheliuzn. Both retinal DA receptors nedrs (binding sites) in myopiccfuck were more than normal Conclusion:The distribuhon of DA receptors in chicken retina are fferent. The increase ofboh DA rmePtors in myopic is relatied to the whction of DA in chick retina in myopia. It indicates that hath DA receptorsInedia retinal DA aIfeCt the develoPInent of myopia.
