水稻受稻瘟病菌诱导的一个基因片段的分离和克隆
ISOLATION AND CLONING OF NOVEL RICE GENE FRAGMENT INDUCED BY Magnaporthe grisea
【摘要】 本研究利用一对抗瘟性近等基因系(near-isogenic lines,NILs)H7R(抗病)和H7S(感病),经稻瘟病菌诱导后提取RNA,进行m RNA 差异显示(differentialdisplay,DD)。在获得多个差异cDNA 片段的基础上,经 PCR 重扩增,Southern 杂交粗筛,Northern 进一步鉴定。其中 25 号片段(克隆RM a25)经序列测定,国际联网查询,证实是受稻瘟病菌诱导的水稻新基因cDNA 片段。
【Abstract】 Total RNA was isolated from a set of rice near isogenic lines H7R(resistant)and H7S (susceptible)that were inoculated by Magnaporthe grisea .Differential display of mRNA was employed to detect the differentially expressed transcripts.After re amplifying the dif ferential cDNA fragments by PCR,Southern and Northern hybridization were performed.Results from sequencing and checking index through Internet proved that clone RMa25 is a novel rice cDNA gene fragment induced by M.grisea.
