组织因子途径抑制因子和生物材料血液相容性关系的体外研究
EFFECT OF TISSUE FACTOR PATHWAY INHIBITOR ON HAEMOCOMPATIBILITY OF POLYMERS: in vitro STUDY
【摘要】 本文试就组织 因子途径抑制因 子( Tissue factor path w ay inhibitor , T F P I) 和生物 材料 血液相 容性 之间的关系进行 了初步的探 讨。本 研究 通过 基因 重组的 方法 制备 了人 重组 T F P I,将 其 涂于 医用聚 乙烯和聚氯乙 烯薄膜材料表面 ,观察 T F P I 涂层对接触血液 的凝血时间、血 液中组织 因子及 凝血因 子 Xa 活性的影 响。结果显示:1实验组( T F P I 处理) 的材料表面 与血液接触后 其凝血时间 比对照 组( 未经 T F P I 处理) 明显延 长,动 态凝 血时 间也 得到 类似 结果;2实 验组 材料 表面 对 接触 血液 中组织因子及凝 血因子 Xa 的活 性有明显的 抑制作 用。上述 结果提 示 T F P I 在 生物 材料 的血 液相 容性改性方面具 有十分广阔的 应用前景。
【Abstract】 Possibillity of applying tissue factor pathway inhibitor(TFPI) to improve haemocompatibility of polymers was investigated. For this purpose, recombinant GST TFPI fusion protein was prepared by purification with glutathione agarose affinity column from E.coli. bacteria which was constructed at our lab through DNA manipulation. Medical grade polyvinylchloride(PVC) and polyethylene(PE) membranes were coated with GST TFPI fusion protein or GST protein by incubating them together at 4℃ for 24 hours, with membranes which were only treated with buffer as controls. The treated or untreated membranes were compared in terms of blood coagulation time and dynamic blood coagulation time occurred on their surfaces due to inhibition of activities of tissue factor and factor Xa. Our results showed that after being coated with GST TFPI, both PVC and PE membrane had an inhibitory effect on the activities of tissue factor and factor Xa in the blood contacted with the membranes when compared with GST coated and control membranes. Our results also showed that GST TFPI fusion protein coating could significantly prolong the blood coagulation time, while GST protein coating had little effect. The inhibition of the activities of tissue factor and factor Xa by TFPI may contribute to the mechanism by which TFPI affects the blood coagulation time. Our study suggested that TFPI may provide a new approach to the improvement of haemocompatibility of biomaterials.
