三株弓形虫速殖子的体外培养研究
A STUDY ON IN VITRO CULTIVATION OF 3 STRAINS OF TOXOPLASMA GONDII TACHYZOITES
【摘要】 目的为探索一种以体外培养方法收获弓形虫速殖子的较适条件。方法以RH、SH1、PP3个分离株的弓形虫速殖子,按105、104、103的接种量分别接种HeLa细胞,调定培养基的pH值分别为8.0、7.0和6.5进行培养。结果发现接种后6d速殖子的累积收获量存在差异性,其中以SE1株收获量大,RH株次之,PP株最少;接种量为105时,收获量大,104组次之,103最少。在培养基pH为6.5时的收获量大,pH7.0次之,pH8.0最少。结论pH6.5的培养条件较适于试验的3株弓形虫速殖子体外增殖,同时,弓形虫的体外增殖也与接种量及不同弓形虫分离株有关
【Abstract】 Aim To investigate the relatively optimal conditions for in vitro cultivation of T.gondii for obtaining a better harvest of tachyzoites.Method 3 isolates,viz.RH、SH1、PP,were cultivted in HeLa cells with 3 different iniital inoculates(10 5、10 4、10 3)and adjusted the medium in 3 pHs(8 0、7 0、6 5).Results It was found that,harvesting at the 6th day after inoculation,there were distinct differences among the cumulative harvest of tachyzoites.Harvest collection in SH1 was the highest versus PP the lowest.When the pH 6 5 was applied,the harvest was higher than that of 7 0,and in 8 0 was the lowest.Similarly,the harvest decreased along with decreases of the initial inoculum,i.e.10 5,the highest and 10 3,the lowest.Conclusion It may be concluded that pH 6 5 is a suitable environment for multiplication of T.gondii tachyzoites in vitro.The amount of the initial inoculum and the strain of isolate difference may also have influence on multiplication of the parasite in vitro.
