gp130分子在树突状细胞上表达及其意义

【摘要】 目的:探讨单核细胞向树突状细胞(DC)分化成熟时,gp130分子在细胞膜上的表达及其意义。方法:人外周血分离得到的单核细胞在含有DC分化成熟所需的各种因子的培养液中分别培养4d和7d,然后用流式细胞术测定DC上gp130分子的表达。结果:gp130分子在单核细胞上低表达,经培养分化成熟成为DC后,DC上的gp130分子表达被上调,特别是在TNFα和抗CD40刺激型单克隆抗体存在下。结论:gp130分子表达于DC前体细胞(单核细胞)并随其分化成熟而增加。

【Abstract】 Objective: To investigate expression of gp130 molecule on dendritic cells (DC) when monocytes differentiated and matured into DC. Methods: Monocytes isolated from human peripheral blood were cultured with different combination of cytokines required for DC differentiation and maturation for 4 days and 7 days respectively, the expression of gp130 molecule on DC was analyzed by cytometry. Results: gp130 molecule was low expression on monocytes, and up regulated expression on DC after monocytes were grown and induced into mature DC, especially in the presence of TNFα and CD40 stimulating monoclonal antibody. Conclusion: gp130 molecule is present on monocytes and is increased on mature DC generated from monocytes.
