弓形虫诊断抗原的亲和层析纯化和分析

【摘要】 应用琼脂糖凝胶Sepharose4B亲和层析分离弓形虫速殖子细胞质抗原(Ps)和弓形虫速殖子膜抗原(Pm),分别获得亲抗体Ps和亲抗体Pm。经双多抗酶联免疫吸附试验(ELISA)夹心法测定,结果表明,Ps的最低检出量为13.3μg/ml,亲抗体Ps为1.5μg/ml,Pm为4.4μg/ml,亲抗体Pm为05μg/ml。作为弓形虫诊断抗原,亲抗体Ps和亲抗体Pm明显优于Ps和Pm,其中又以亲抗体Pm为最优。

【Abstract】 Cytoplasm antigen (Ps) and membrane antigen (Pm) of toxoplasm tachyzoites were parted by Sepharose 4B aflinity chromatography. Parental antibody Ps and parental antibody Pm were gained and determinated by ELISA.The minimal detectable concentration of Ps,parental antibody Ps,Pm and parental antibody Pm was 13.3 g/ml,1.5 g/ml,4.4 g/ml and 0.5 g/ml,respectively.As diagnostic antigen of toxoplasm,parental antibody Ps and parental antibody Pm were significantly superior to Ps and Pm,and parental antibody Pm was the best among them.
