PCR/DNA探针检测系统用于间日疟原虫检出及基因型检测的研究
STUDIES ON DETECTION AND GENOTYPE OF PLASMODIUM VIVAX BY METHOD OF PCR/DNA PROBE DETECTION SYSTEM
【摘要】 根据间日疟原虫环子孢子表面蛋白(CSP)基因序列研制了PCR/DNA探针检测系统。试验证明该检测方法对间日疟原虫特异,检出原虫密度为0.2个/μl,病人阳性检出率为96.2%。试验中对不同PCR操作过程及试验样本不同处理方法的检测结果进行了比较。在样本的原虫基因型检测分析中,基因优势型PV210阳性率为88.5%,基因变异型PV247阳性率为11.5%,混合型阳性率为3.8%,基因变异型均出现在云南省采集的血样中
【Abstract】 Based on the gene sequence of circumsporozoite protein(CSP) of Plasmodium vivax , the PCR/DNA probe detection system was prepared. The experiment showed that this detection system was specific to P. vivax , and its detecting sensitivity was 0.2 parasite/μl. The results in different procedures of PCR operation and in different methods of handling experimental samples were compared. The two kinds of CSP genotype of P. vivax were also identified by the detection system. The positive rates were 88.5% for PV 210 CSP gene form, 11.5% for PV 247 and 6.3% for mixed form respectively. Further analysis of the results showed that the PV 247 form only appeared in samples from Yunnan Province.
