再生障碍性贫血患者骨髓造血干/祖细胞c-kit受体表达
Expression of c kit receptor on the hemopoietic stem/precursor cells of aplastic anemia patients
【摘要】 目的 观察再生障碍性贫血(再障) 患者骨髓造血干/祖细胞ckit 受体(CD117) 的表达,以了解再障造血功能衰竭与ckit 受体水平间的关系。方法 采用免疫荧光法及流式细胞技术检测20例再障患者骨髓单个核细胞(MNC) 和CD34+ 细胞CD117 的表达水平。结果 ①MNCCD117 表达率慢性再障(CAA) 组为(2 .08 ±1.96) % 、重型再障(SAA)组为(0.89 ±0.32) % 、正常对照组和增生性贫血组分别为(0 .45 ±0.16) % 和(0.67±0 .39) % ,CAA 组和SAA 组明显高于正常对照组和增生性贫血组( P<0-05) ;②CD34 CD117双阳性细胞百分率,CAA 组、SAA 组、正常对照组和增生性贫血组分别为(0 .87 ±0-57) % 、(0.65±0.41)% 、(0.41±0 .15)% 和(0.46±0.26)% ,CAA 组和SAA 组明显高于正常对照组和增生性贫血组( P< 0.05);③CD34+ 细胞群CD117 的表达,CAA 组、SAA 组、正常对照组和增生性贫血组分别为(71 .6±12.4)% 、(88.9 ±23.5) % 、(46 .3±9 .7) % 和(41.8 ±?
【Abstract】 Objective To explore the expression of c kit receptor(CD 117 ) on the hemopoietic stem/precursor cells of aplastic anemia patients and its role in the genesis of aplastic hematopoiesis failure.Methods c kit expression on the bone marrow mononuclear cells(MNC) and CD 34 positive cells from 11 chronic aplastic anemia(CAA),9 severe aplastic anemia(SAA),10 non aplastic anemia(NAA) patients (5 iron deficiency anemia and 5 autoimmune hemolytic anemia) and 5 healthy controls was measured with flow cytometric immunofluorescence assay and anti CD 117 monoclonal antibody.Results The CD 117 positive rates of MNC in CAA,SAA,NAA and control groups were (2.08±1.96)%, (0.89±0.32)%, (0.67±0.39)% and (0.45±0.16)%,respectively,and the rates were significantly higher in CAA and SAA groups than in NAA and control groups(P<0.05).The CD 117 and CD 34 double positive cells of MNC in CAA,SAA,NAA and control groups were (0.87±0.57)%,(0.65±0 41)%,(0.46±0.26)% and (0.41±0.15)%,respectively,and were significantly higher in CAA and SAA groups than in NAA and control groups (P<0.05).The CD 117 positive cells of CD 34 positive cells were also higher in CAA[(71.6±12.4)%] and SAA[(88.9±23.5)%] groups than in NAA[(41.8±10.3)%] and control[(46 3±9.7)%] groups (P<0.01).Conclusion Hemopoietic stem/precursor cells of aplastic anemia patients expressed more c kit receptors than those of NAA patients and healthy controls.The hematopoiesis failure occurred in aplastic anemia is not attributed to low expression of c kit receptor on the hemopoietic stem/precursor cells.
