γδT细胞及其培养上清成份对骨髓造血细胞集落形成的影响
Study on modulatory role of γδT cell and its supernatant for hematopoiesis
【摘要】 目的 研究γδT 细胞及其培养上清成份对骨髓造血细胞集落形成的影响。方法 运用体外半固体造血集落培养观察了人γδT 细胞及其分泌上清对自体骨髓造血集落(CFUC) 形成的影响。结果 γδT 细胞及其培养上清能刺激自体骨髓造血集落的形成;其培养上清中可检出IFNγ、TNF、IL8 等细胞因子。结论 γδT 细胞能分泌一系列具有不同效应的造血调控因子,但其共同的作用结果则具有一定的造血刺激效应,可能成为一种新型的CTL 用于肿瘤的过继免疫疗法。
【Abstract】 Objective To study the modulatory role of the γδT cell form human peripheral blood on growth of autologous bone marrow progenitors (CFU C) and the levels of the cytokines in γδT cell supernatant. Methods By making use of cultures for colonies of hematopoiesis with semi solid medium, the effects of human γδT cells and its supernatant on the formation of CFU C were observed. Results It showed that γδT cell or its supernatant could stimulate CFU C formation in the system without external cytokines, although the CFU C numbers was lower. Meanwhile, high levels of cytokines were detected in γδT cell supernatant. Conclusion These results strongly support the implication possibility of γδT cell on tumor immunotherapy and bone marrow purification.
