人胰腺癌细胞血管生成过程的观察及意义
Observation of the angiogenesis process of human pancreatic cancer cells and its clinicai implication
【摘要】 目的探讨人胰腺癌血管生成方式和时间,分析其临床意义,为寻求抗人胰腺癌血管生成药物做准备。方法用SCID鼠制做皮肤血管室模型,血管室内接种人胰腺癌细胞株,活体显微镜下连续观察2周,录制图像,使用图像分析软件比较血管面积、密度及直径的变化。结果1周内新生血管基本完成,2周内形成肉眼可见的微小肿瘤;血管密度和面积的变化显著(P<0.001,P<001);血管直径的变化差异无显著性(P>0.05)。结论人胰腺癌细胞可诱导周围正常血管内皮细胞增生形成肿瘤血管;胰腺癌术后的辅助治疗应开始于术后2周以内;血管密度可作为估计胰腺癌恶性程度高低的生物学指标。
【Abstract】 Objective To investigate the angiogenesis process of human pancreatic cancer cells(HPCC) and its clinical implication. Methods Cancer cells were implanted into the dorsal skinfod charnberon SCID mice and angiogenesis process was continuously observed with introvital videomicrosc0pic system fortwo weeks. Imaging analysis software was used to compare the changes in vascular area (VA), vascular density (VD) and vascular diameter. Results HPCC couId form their own new vasculature from surrounding normal microvessels in one week, and visible microtumors in the chamber could be found in two weeks. Thechanges in VD and VA were significant as compared with control group (P<0. 001, P< 0.01 respectively).Condusions HPCC could induce angiogenic agents which stimulate the proiferation of surmunding endothelial cells and form new tumor vasculature. Antiangiogenic therapy might be used to treat this disease. Postoperative adjuvant treatment should be given as earlier as peibIe. VD might be used as a gcod indicator of themalignant extent of human pancreatic cancer.
