【摘要】 目的探讨中药“热毒清”抗人巨细胞病毒（ＨＣＭＶ）的实验和临床效果及作用机制。方法采用定量ＲＴＰＣＲ方法，检测中药“热毒清”干预后感染细胞ＨＣＭＶ晚期ｍＲＮＡ的表达水平，观察感染细胞的病变进展，并在体外实验的基础上，将“热毒清”应用于ＨＣＭＶ活动性感染孕妇的临床治疗，测定治疗前后血清肿瘤坏死因子α（ＴＮＦα）和白细胞介素２（ＩＬ２）的水平。结果“热毒清”对ＨＣＭＶ晚期ｍＲＮＡ以及ＨＣＭＶ所致细胞病变有明显的抑制作用。临床观察发现，ＨＣＭＶＩｇＭ阳性孕妇和育龄妇女ＴＮＦα较对照组显著升高，而ＩＬ２则显著下降；“热毒清”治疗后ＴＮＦα和ＩＬ２迅速恢复至正常水平，ＨＣＭＶＩｇＭ转阴率为７２９％，显著高于未治疗组１３６％的自然转阴率。结论血清ＴＮＦα水平与体内ＨＣＭＶ的活动状态有关，ＨＣＭＶ对机体有免疫损伤效应，“热毒清”能够提高机体的免疫功能，在体内外对ＨＣＭＶ均有显著的抑制作用，是治疗ＨＣＭＶ活动性感染，尤其是孕期感染安全有效的药物。更多还原

【Abstract】 Objective To study the theurapeutic effect of Chinese medicine ReDuQing on HCMV. Methods The expression of the HCMV latemRNA in infected cells was measured by quantitative RTPCR and the development of cytopathic effect (CPE) caused by HCMV was observed before and after the treatment of Chinese medicine ReDuQing. On the basis of its antiHCMV activity study in vitro, ReDuQing was applied to the childbearing aged women and to pregnant women who were HCMVIgM positive which indicated the active HCMV infection, meanwhile the serum levels of tumor necrosis factoralpha (TNF) and interleukin2 (IL2) were measured before and after the treatment by the ELISA method. Results The results showed that the ReDuQing had strong inhibition effect both on HCMVmRNA and CPE. The serum level of the TNF increased and IL2 decreased significantly in HCMVIgM positive cases when compared with normal women. After being treated with ReDuQing, the serum TNF and IL2 in patients returned to normal level, and 72.9% of the HCMVIgM positive cases turned negative which was significantly higher than that (13.6%) in control group. Conclusion It was suggested that serum TNF level is closely related to the activity of HCMV which has harmful effect on immune system. ReDuQing can enhance immunity of the body and has an obvious inhibition on HCMV in Vitro and Vivo, and can thus serve as safe and effective medicine for treatment of active HCMV infection, especially for the pregnant women.更多还原
