蛛网膜下腔移植转脑啡肽基因细胞用于治疗神经性疼痛

【摘要】 目的评价蛛网膜下腔移植人前脑啡肽原基因(hppe)修饰细胞用于神经性疼痛治疗的效果。方法采用可吸收羊肠线结扎40只大鼠单侧坐骨神经,制作慢性神经痛模型(成功34只)。鼠成纤维细胞系 NIH/3T3细胞在脂质体介导下转染hppe基因,转化细胞记3T3/hENK。 34只慢性神经痛模型 SD大鼠随机分成 2组:腰段蛛网膜下腔植入 NIH/3T3细胞为对照组,腰段蛛网膜下腔植入3T3/hENK细胞为实验组。观察动物的电、热痛阈和自发痛行为的改变,以及腹腔注射纳洛酮对细胞镇痛效应的影响。结果实验组大鼠的疼痛明显减轻,电痛阈由(0.28±0.05) mA升至(0.68±0.18)mA,热痛阈由(12.22±2.09)秒升至(27.08±6.4)秒(P<0.05),而对照组的热痛阈和电痛阈无显著变化(P>0.05);与对照组相比,实验组自发痛行为明显改善。实验组的镇痛效应可被纳洛酮拮抗。结论蛛网膜下腔移植hppe基因修饰细胞可有效改善神经性疼痛,其镇痛效应是通过阿片受体介导的。

【Abstract】 Objective To evaluate the efficacy of subarachnoid implantation of somatic cell line genetically modified with human preproenkephalin gene on neuropathic pain. Methods The chronic neuropathic pain was induced with tying off sciatic nerve of rat. The NIH/3T3 cells of rats, the fibroblast like cell line, were trans feeted with a human preproenkephalin gene to be transformed into 3T3/hENK cells. Thirty-three Sprague-Dawley rats with the unilateral ischialgia, were randomly divided into two groups: group test with subarachroid implantation of 3T3/hENK cells,group control with subarachroid implantation of NIH/3T3 cells. Such parameters were tested as electrical/ thermal hyperalgesia, behavioral indications of spontaneous pain and the effects of intraperitoneal naloxone on the analgesic efficacy. Results in group test the electrical and thermal threshold increased from (0. 2810. 05) mA and (12. 2212. 09) s to (0. 6810. 18) mA and (27.08±6.4) s, respectively (P<0. 05), but in group control remained unchanged (P>0. 05). The behavioral indications of spontaneous pain decreased in group test as compared with those in group control. These efficacies were reversed by intraperitoneal naloxone in group test. Conclusion These results establish the analgesic efficacy of intrathecal somatic cells trans feeted with enkephalin gene in a chronic pain model of nerve injury, which is conducted via opiate receptors.
