Fas表达水平与抗Fas治疗生物学效应的关系
Anti Fas on bladder carcinoma cell line EJ:level of Fas is a predictive marker of biological responsiveness
【摘要】 目的 研究细胞表面Fas 表达水平与抗Fas 治疗生物学效应之间的关系。 方法 将低表达Fas 的膀胱癌细胞系EJ细胞通过脂质体介导的方法转染Fas 基因,并用流式细胞直接免疫荧光术检测Fas 的表达;采用鼠抗人Fas 单克隆抗体DX2 IgG1κ诱导转染Fas 基因的和未转染Fas 的EJ细胞的细胞凋亡作用,并用碘化丙啶染色流式细胞术检测。 结果 转染Fas 基因后的EJ细胞Fas分子表达明显增加,Fas 分子表达率从26 .11 % 增加到68.69% ;鼠抗人Fas 单克隆抗体DX2 IgG1κ诱导的转Fas 基因的EJ细胞凋亡数明显增多,达66 .3% ,而未转基因的EJ细胞凋亡较少,仅7 .4% 。 结论 人类恶性肿瘤细胞系对Fas 抗体触发凋亡信号的敏感性主要依赖于细胞表面Fas 的表达水平。
【Abstract】 Objective To investigate the relationship between expression level of Fas and anti Fas biological responsiveness. Methods With direct immunofluorescence flow cytometry, expression of Fas was detected in bladder carcinoma cell line EJ transfected and untransfected with Fas gene.Apoptosis was induced with anti Fas antibody DX2 IgG1κ and measured by flow cytometry. Results Expression of Fas was increased significantly, positive cell number increased from 26.11% to 68.69%; Apoptosis induced with anti Fas antibody DX2 IgG1κ (in bladder carcinoma cell line EJ transfected with Fas gene) reached 66.3%,significantly highier than that in bladder carcinoma cell line EJ untransfected(7.4%). Conclusions The sensitivity of human malignant cell to Fas mediated apoptosis is mainly dependent upon the expression of Fas.
