雌激素与孕激素对卵巢癌耐药细胞系OVCAR-3生长及耐药的影响
Effects of Estradiol and Medroxyprogesterone on the Growth and Doxorubinresistance of Drug-resistant Human Epithelial Ovarian Cancer Cell the OVCAR-3
【摘要】 目的研究雌、孕激素对卵巢癌耐药细胞系OVCAR-3生长及对阿霉素(ADM)耐药的影响。方法用四甲基偶氮唑蓝(MTT)快速比色法,计算细胞生存率,以比较不同浓度雌、孕激素对细胞生长的影响,及雌、孕激素与异搏定对卵巢癌耐药细胞系OVCAR-3耐药性的影响。结果高浓度的苯甲酸雌二醇(E2,10.00~100.00μmol/L)和安宫黄体酮(MPA,100.00μmol/L)可抑制OVCAR-3细胞的生长,且二者联合抑制作用增强;而较低浓度的E2(0.01-1.00μmol/L和MPA(0.01~10.00μmol/L)对该细胞的生长无明显影响;异搏定可增加ADM对OVCAR-3细胞的抑制作用,即逆转该细胞对ADM的耐药;E2或MPA均可增加ADM对OVCAR-3细胞的抑制作用,即逆转该细胞对ADM的耐药,且其作用强于异搏定。结论超生理浓度的雌、孕激素可抑制OVCAR-3细胞的生长,而较低浓度的雌、孕激素对其生长无影响;雌、孕激素可逆转OVCAR-3细胞对ADM的耐药,且作用强于异搏定,可望成为临床化疗的辅助用药。
【Abstract】 Objective To study the effects of estradiol (E2) and medroxyprogesterone acetate (MPA) on the growth of drug resistant human epithelial ovarian cancer cell line. Methods OVCAR-3 cells, treated by different concentrations of E2, MPA and verapamil, were examined for their growth inhiitions with methyl thiazlyl tetrazolium (MTT, a tetrazolium dye) rapid photocolorimetric assay. Results OVCAR-3 cell growth was inhibted by E2 at high concentrations (10.00- 100.00 μmol/L)and by MPA at 100.00μmol/L, while lower concentrations of E2(0.01-1 .00 μmol/L) and MPA(0.01-10.00 μmol/L) had no effect on OVCAR-3 cell growth, neither stimulation nor inhibihon. The effect of E2 combined with MPA was more Obvious than either one used separately.E2 and MPA in combination with doxorubin showed a synergistic inhibition effect on OVCAR-3 cell growth, which suggesed that both of them could reverse the doxorubin resistance of OVCAR-3 cell line. The inhidition effect of Verapamil combined with doxorubin was obviously weaker than that of E2 and MPA. Conclusions E2 and MPA could reverse the doxorubin resistance of OVCAR-3 cell line and their inhibhon were more obvious than the of verapamil, which Suggested that E2 and MPA may be used clinically for adjuvant chemotherapy.
