卵巢癌抗独特型抗体融合蛋白细胞免疫功能的体外实验
In vitro Study of 6B11GM to Induce Cellular Immunity in Patients with Ovarian Carcinoma
【摘要】 目的探讨卵巢癌抗独特型单链抗体(6B11scFV)与人粒细胞-巨噬细胞集落刺激因子(hGM-CSF)形成的融合蛋白(6B11GM)作为肿瘤疫苗的可能性。方法从卵巢癌病人外周血中分离得到单个核细胞、T淋巴细胞和作为抗原呈递细胞的单核细胞,同时从病人腹水中分离得到癌细胞,分别以卵巢癌抗独特型抗体的全抗体(6B11)、6B11GM、6B11+hGM-CSF、hGM-CSF进行体外T淋巴细胞增殖实验和自身肿瘤细胞杀伤实验。并对6B11和6B11GM的作用进行比较。结果6B11GM能诱导抗原特异性T淋巴细胞增殖,其作用优于6B11;6B11GM能增加外周血单个核细胞对自身肿瘤细胞的杀伤作用。结论6B11GM能诱导抗原特异性的细胞免疫反应,为卵巢癌的主动免疫治疗提供新的方法。
【Abstract】 Objective In order to explore the possibility of ovarian carcinoma anti-idiotypic single chain antibody (6B11scFV) and human granueocyte-macrophag colony stimulating factor facto (hGM-CSF) fusion protein 6B11GM as tumor vaccine. Methode Mononuclear cells, T cells and monocyte used as antigen presenting cell (APC) were separated from peripheral blood, and tumor cells were obtained from ascitic fluid in the same patients.In vitro T cell proliferation and autologous tumor cell cytotoxicity assay were apphed.Results 6B11GM could indue T cell proliferation in vitro which was antigen speific. Cytotoxicity of mononuclear cells to autologous tumor cells increased after primed with 6B11GM.Conclusion 6B11GM could induce efficient cellular immunity in patients with ovarian carcinoma,hopefully may be used for ovarian carcinoma vaccine.
