【摘要】 目的：研究非小细胞肺癌患者单次低剂量口服鬼臼乙叉苷软胶囊药物动力学，并设计较为合理的给药方案。方法：ＨＰＬＣ法测定血药浓度，ＭＣＰＫＰ程序处理数据。结果：其药动学参数符合一室开放模型，Ｋａ 为１ ．８ ±２．２ ｈ－ １ ，Ｋ 为０．１９ ±０．０２ ｈ－１ ，Ｔ１／２Ｋａ１ ．０±０．８ ｈ，Ｔ１／２Ｋ为３．６±０．４ ｈ，Ｔｍａｘ 为２ ．４ ±１ ．２ ｈ，Ｃｍａｘ 为３ ．３±１．１ ｍｇ·Ｌ－１ ，ＡＵＣ为２６ ．１±６ ．２ ｍｇ·ｈ·Ｌ－１ 。药动学参数经血管外多次重复给药方案设计程序处理。结论：２５ ｍｇ·ｍ － ２ ，每天２ 次的治疗方案较为合理。更多还原

【Abstract】 OBJECTIVE:To investigate the pharmacokinetics of oral low dose etoposide in 8 patients non samll cell lung cancer and to suggest reasonable regimen.METHODS:High performance liquid chromatography was used to determine received orally 50 mg·m -2 single etoposide in plasma.MCPKP program was used for the calculation of pharmacokinetics parameter.RESULTS:The pharmacokinetics was in accord with single compartment model and K a was 1.8 ± 2.2 h -1 , K was 0.19 ± 0.02 h -1 , T 1/2Ka was 1.0 ± 0.8 h, T 1/2K was 3.6 ± 0.4 h, T max was 2.4 ± 1.2 h, C max was 3.3 ± 1.1 mg·L -1 , AUC was 26.1 ± 6.2 mg·h·L -1 .Based on the pharmacokinetics of etoposide after single oral dosing,the plasma levels were predicted on chronic daily dose(50 mg·m -2 ,qd and 25 mg·m -2 ,bid) maintenance of daily plasma concentration 1 to 3 mg·L -1 would be about 8 h for dose 50 mg·m -2 ,qd and 12 h for dose 25 mg·m -2 ,bid.CONCLUSION:The administration of oral etoposide 25 mg·m -2 ,two times daily as reasonable are safe regimen was recommended for the treatment of non small cell lung cancer.更多还原
