不同栽培居群红花的孢粉特征、同工酶谱及化学成分含量

【摘要】 目的:探讨红花的种内变异,为不同栽培居群红花的鉴别及优良栽培居群的选育提供依据。方法:在对不同栽培居群红花进行引种栽培的基础上,扫描电镜观察花粉粒特征,垂直平板电泳法分析细胞色素氧化酶同工酶谱带,UV、HPLC 测定黄色素和腺苷含量。结果:红花的孢粉特征及细胞色素氧化酶谱带特征既有种一级的共同特征,又反映了种内存在一定的变异;不同栽培居群化学成分含量之间差异的规律性与在原产地栽培时基本一致。结论:红花苞叶细胞色素氧化酶谱带可以从分子水平鉴别不同的栽培居群;红花中化学成分含量的高低,主要与遗传因素有关,同时,也受环境条件的影响。为红花的引种栽培以及优良栽培居群的选育提供依据。

【Abstract】 OBJECTIVE: To investigate the intraspecific variance of Safflower, provide the basis for distinguishing the different cultivated population and breeding of fine cultivated population. METHODS: Based on the introduction and cultivation of four cultivated population, the pollen features were observed by scanning electron microscope,the bands of cytochrome oxidase were analysed by vertical slab electrophoresis,the contents of safflower yellow, polysaccharide and adenosine were determined by UV, HPLC.RESULTS:The pollen features of Safflower and bands of cytochrome oxidase had not only specific features, but also reflected the intraspecific variance of Safflower. The rules of the difference of the contents of different cultivated population were basically consistent with that in original localities.CONCLUSION:The bands of cytochrome oxidase in involucre of Safflower could distinguish different cultivated population at the molecular level.The contents of chemical components in Safflower were mainly related to hereditary factors,and also were affected by the ecological environment.The results here provided basic comprehension for introducing, cultivating and breeding the fine cultivated population of Safflower.
