高频电刺激丘脑底核对帕金森病大鼠旋转行为及纹状体多巴胺和兴奋性氨基酸的影响

【摘要】 目的 观察高频电刺激丘脑底核对帕金森病( Parkinson's disease ,PD)大鼠旋转行为及纹状体神经递质的影响,探讨电刺激治疗PD的作用机理。方法 采用6-OHDA建立纹状体毁损PD大鼠模型, 观察高频电刺激丘脑底核后PD鼠旋转行为的变化, 并同时检测纹状体中多巴胺和兴奋性氨基酸的含量。结果 电刺激丘脑底核后,改善了PD大鼠的旋转行为,纹状体中多巴胺含量明显增加( P< 0.01),谷氨酸含量明显减少(P< 0.01),天门冬氨酸则无明显改变( P> 0.05)。结论 高频电刺激丘脑底核促进了双侧纹状体多巴胺的释放, 抑制了同侧谷氨酸的释放,从而有助于PD症状的改善。

【Abstract】 Objective To observe the effects of rotation behaviour and the contents of neurotransmitter in the striatum of Parkinson’s disease( PD) rats by the high frequency stimulation to the subthalamic nuclus . Method The model of PD was founded by injecting 6 OHDA into the right striatum of rats .After the stimulation to the STN, we observed the effects of the rotation behaviour of the rats to determine the content of the dopamine(DA) and excitatory amino acids (EAAs) in the striatum . Results After the stimulation ,the rotating behaviour induced by apomorphine and improved distinctly, and DA content in the striatum increased (P<0.01), and Glu content reduced significantly ( P<0. 05).Content of the ASP had no change significantly. Conclusion These results indicate that the high frequency stimulation to STN can elicit DA’s releasing in the bilateral striatum, and block the releasing of Glu in the ipsilateral striatum .These may be benificial for treatment of PD.
