中枢神经递质γ-氨基丁酸与其合成阻断剂对大鼠血糖和胰岛素的影响

【摘要】 自的观察中枢神经递质γ-氨基丁酸与其合成阻断剂对大鼠血糖和胰岛素的影响。方法Wistar大鼠侧脑室注射不同剂量的神经递质之一γ-氨基丁酸(camma-aminobutyricacid,GABA)及GABA谷氨酸脱羧酶抑制剂3-巯基丙酸(3-mercaptopropionicacid,3-MP)后,观察不同时间大鼠血糖及血清胰岛素的改变,并设对照组注射生理盐水进行比较。结果注射GABA后,血糖较对照组明显升高,同时胰岛素含量明显降低,且这些变化与注射GABA的含量呈剂一效关系。注射GABA谷氨酸脱羧酶抑制剂3-MP后,血糖较对照组下降。结论初步证明神经递质对糖代谢及糖尿病患者血糖变化存在着重要的影响。

【Abstract】 Objective To study the influences of GABA and 3-MP on the blood glucose concentrations and insulin levels in normal Wistar rats. Methods Three groups of Wistar rats (male, body weight 200-300g) were given GABA (10μg), 3-MP(50μg),and NS(10μl) individually by nitracererbroventricular injecticns. Blood samples were taken from the orbital veins.Blood glucose was estimated by glucose oxidase method, and insulin was measured by RIA. Results In the GABA group, the blood glucose increased after 10’ after administration and were getting higher and higher with the peak at 90’;The insulin levels were lower correspondingly. In the 3-MP group,the glucose concentrations were getting lower and lower with corresponding insulin levels increased. Conclusion GABA plays an important role on the homeostasis of blood glucose and insulin activity. It may be of pathogenetic significance in diabetes mellitus.
