血管紧张素Ⅰ转化酶基因多态和2型糖尿病肾病相关性研究
A study on the association of an insertion/deletion (I/D) polymorphism of angiotensin Ⅰ converting enzyme(ACE) gene with Chinese type 2 DM diabetics with nephropathy
【摘要】 目的研究血管紧张素Ⅰ转化酶(ACE)基因多态和2型糖尿病肾病(DN)的关系。方法用PCR技术检测102例中国汉族2型糖尿病患者ACE基因1/D多态基因型。结果DN组(n=36)和无DN组(n=49)相比,D型等位基因和DD基因型频率升高(分别为59.7%vs38.8%,X2=7.30;33.3%vs12.2%,X2=5.53),有显著性差异(P<0.05)。病程≤5年合并DN组(n=11)与病程>5年无DN组(n=20)相比,DD基因型频率增高(45.5%vs20.0%,X2=4.27,P<0.05)。标化多元逐步回归分析发现依次进入回归方程的是年龄、果糖胺和病程,而基因型未进入方程。结论ACE基因I/D多态和DN有相关性,和某些患者早期发展为DN有关,但在DN的发生发展中是微效基因。
【Abstract】 Objective To study the association of an insertion/deletion (I/D) polymorphism of angiotensin Iconverting enzyme (ACE) gene with diabetic nephropathy (DN). Methods PCR technique was used to determine theACE gene genotypes in 102 cases of Chinese type 2 DM patients. Results in comparison of DN group with non--DNgroup it was found that there was an association of D type allele with DN (X’~ 7’ 30, P <0. 05),and DD genotype wasmore frequent in DN group(X’ = 5’ 53’ P <0. 05). Also, DD genotype was prevalent in DN group of short duration (n= 11 ) than non--DN patients with long duration (n ~ 20) (X’ ~ 4. 27, P < 0. 05). By standard multiple stepwiseregression, it was found that l/D polymorphism of ACE gene could not enter the regression equation. ConclusionPatients carrying DD genotype and D type allele are susceptible to DN. ACE gene polymorphism is associated with earlyoccurrence and development of DN in some patients, but it is a microeffective gene.
