【摘要】 目的　研究维拉帕米（ 异博定，钙离子阻滞剂）、双嘧达莫（ 潘生丁） 、丹参、黄体酮、环孢菌素Ａ、他莫昔芬等药物对人卵巢癌细胞系（ 具有多药耐药的特征） 顺铂（ＤＤＰ） 耐药性的逆转作用。　方法　用ＭＴＴ 法测定上述药物对Ａ２７８０／ＤＤＰ耐药细胞的细胞毒性作用，用高效液相色谱仪（ＨＰＬＣ） 测定细胞内ＤＤＰ 含量。　结果　除丹参以外的其他５ 种药物，能不同程度地逆转耐药细胞对顺铂的耐药性；可增加耐药细胞内顺铂的药物浓度。　结论　耐药细胞逆转作用主要是由于这些药物使耐药细胞内对顺铂解毒系统功能降低，致使细胞内药物浓度增加，与多药耐药基因（ ＭＤＲ１） 关系不大。更多还原

【Abstract】 To investigate the reversional effect on cisplatin(DDP) resistance of verapamil,(a calcium ion channel blocker), dipyridamole(persantin), danshen, progesterone,cyclosporin A and tamoxifen with A2780/DDP human ovarian cancer lines (with multidrug resistance characteristies).METHODS Cytotoxic activity of these drugs against the A2780/DDP cells were observed by MTT method. The concertration of DDP in A2780/DDP cells was determined by high performance liquid chromatographic (HPLC) assay.RESULTS The five drugs (except dansan )reversed the resistance of A2780/DDP cells to DDP in different extent and increased the concentration of DDP in A2780/DDP cells.CONCLUSIONS The reversal mechanism of these drugs may be associated with decreasing detoxification of the cells to DDP,increasing cellular accumulation of DDP and without relation to MDR1.更多还原
