纸血片标本叶酸检测方法的评价研究

【摘要】 目的:纸血片标本用于叶酸营养状况评价是一种新的研究方法,本研究对纸血片标本叶酸微生物学检测方法中叶酸洗脱效率、重复性、贮存条件等进行了初步探讨。结果:纸血片标本叶酸稳定性较好,4℃下贮存3周约有10%叶酸失活;室温下贮存4周叶酸保存率仍在60%以上,避光贮存可降低叶酸损失率;反复冻融对叶酸无明显影响;该检测方法重复性好,批内及批间差异分别<5%,<9%。74名健康非孕妇女叶酸检测结果与放免法红细胞叶酸检测结果呈正相关(r=0.79,P<0.001)。结论:该方法较适用于人群叶酸营养状况的描述性研究及叶酸增补剂服药依从性研究。

【Abstract】 Objective: The folate assay using dried blood spots (DBS) is a new method for folate status screening. We tested the extract efficiency of DBS folate, and studied its stability. Results: The results showed that the folate was stable in DBS. The folate in DBS stored at 4℃ for 3 weeks was lost about 10%, and stored at ambient temperature for 4 weeks, retained above 60%, and decreased slowly after freezing and thawing for 6 times. The storage of DBS in dark container was helpful for folate stability at room temperature. The microbiological assay for the DBS folate had a high precision. The intra and inter run reproducibilities were <5% and <9%, respectively. The mean hemoglobin folate of the non pregnant healthy women (n=74) was 2075±866pmol/L. The results of DBS folate were correlated well with that of radioassay. Conclusion: The DBS is suitable for the descriptive study or pill taking compliance study of folate in the field.
