【Abstract】 The three α 1 adrenoceptor(α 1 AR) subtypes were compared in renal arteries from 12 month old spontaneously hypertensive rats(SHR) and aged matched WKY rats using in vitro contractile studies and RNase protection assays. Contractile studies showed that norepinephrine (NE) induced contraction, including pD 2 values and maximum contractions, was not significantly different between WKY rats and SHR; pA 2 values of α 1A selective antagonists WB4101 and sertindole were increased from 9.08±0.09 and 8.58±0.13 in WKY rats to 9.50±0.24 and 8.94±0.20 in SHR, respectively; whereas pA 2 value of α 1D selective antagonist BMY7378 was not significantly different between WKY rats and SHR (7.59±0.29 vs 7.23±0.27). The results of RNase protection assays showed that mRNA levels for α 1A , α 1B and α 1D AR subtypes were not significantly different between WKY rats and SHR. The results suggest that the potency of NE in renal artery be not significantly different between WKY rat and SHR, but α 1A subtype make a more contribution to NE induced responses in renal artery from SHR than from WKY rats. The functional change may result from the change in receptor protein and/or post receptor signal transduction.
